Oxytocin Modulation Following LI4 and SP6 Acupuncture in Post-Cesarean Section Women: A Randomized Controlled Trial

Abstract

Introduction Cesarean section surgery causes alterations in tissue continuity, resulting in painful trauma, which can pose challenges for lactation and early mobilization, thereby affecting the postpartum uterine involution process. Oxytocin plays a crucial role in stimulating uterine contractions and postpartum lactation. Objective: This study aims to investigate the effectiveness of acupuncture stimulation at LI4 and SP6 points in stimulating the release of oxytocin hormone in postpartum Cesarean section cases.

Methods A randomized controlled group study included 72 postpartum Cesarean section participants from February to May 2024. A random sampling technique was employed to allocate subjects into intervention and control groups using a lottery method. The intervention group received electroacupuncture at LI4 and SP6 points, while the control group received no intervention. Selected respondents met the inclusion criteria and provided signed informed consent forms. Oxytocin levels were analyzed using ELISA testing,

Results Baseline oxytocin levels were comparable among groups (p=0.710). Following intervention, oxytocin levels increased significantly in all acupuncture groups compared with the control group (p<0.001). The combined LI4+SP6 group showed the greatest increase, from 41.44±11.50 pg/mL to 95.34±8.32 pg/mL (mean difference: 53.90±13.24 pg/mL). In comparison, oxytocin levels increased by 19.02±8.90 pg/mL in the LI4 group, 22.06±8.59 pg/mL in the SP6 group, and 4.66±6.54 pg/mL in the control group.

Conclusion The results of this study indicate that acupuncture intervention in postpartum Cesarean section cases can stimulate oxytocin hormone release, which may aid in lactation processes and promote uterine contractions during uterine involution
